Dopamine neurons in ventral mesencephalon: interactions with glia and locus coeruleus

Parkinson’s disease is a progressive neurodegenerative disorder, described as a exhaustion of the dopamine neurons in the substantia nigra. The reason behind the disease is still not known but age, oxidative stress, and neuroinflammation are a few of the features involved in the destruction. Additionally, considerable cell death of noradrenergic neurons occurs in the locus coeruleus (LC). Noradrenaline has long been recommended to defend the dopamine neurons from oxidative stress and neuroinflammation. The key cure for Parkinson’s disease is Levo-dopa, despite the fact that significant negative effects occur because of this therapy. Hence, grafting fetal ventral mesencephalic (VM) tissue into the adult striatum has been examined as a replacement therapy for Parkinsons’s disease. Having said that, the survival of the grafted neurons is restricted, and the dopamine-denervated striatum doesn’t become fully reinnervated. Thus, elucidating factors that improve dopamine nerve fiber creation and/or survival of the grafted neurons is very important.To research dopamine nerve fiber formation and also the connections with glial cells, organotypic VM tissue cultures were used. Two morphologically distinct nerve fiber outgrowths from the tissue slice were observed. Nerve fibers were initially formed in the absence of migrating astrocytes, however thin vimentin-positive astrocytic processes were discovered within the same area.
